Alcoholism drug linked to dramatic weight loss in obesity study – SlashGear

A new study has linked an old school drug used to treat alcoholism with rapid weight loss and protective effects in obese mice, hinting at a potential new treatment for obesity. The research comes from the National Institutes of Healths National Institute on Aging, which found that mice given the medication disulfiram lost weight much faster than mice that only went on a diet.

Disulfiram is a drug that has been used to treat alcohol use disorder for more than five decades, according to NIH, which found that it may also be able to protect against metabolic damage associated with obesity while helping return the subject to a healthy weight. The study involved both male and female mice; it is described as a basic research finding.

When it comes to alcoholism, disulfiram works by causing the patient to feel nauseous if they consume alcohol. Researchers became interested in studying the drugs potential use in cases of metabolic disorders and obesity due to the class of drugs reputation for treating rats with type-2 diabetes.

The study involved a total of four groups of lab mice that were fattened up over the course of 12 weeks: one fed a high-fat diet, another fed a high-fat diet alongside a low dose of disulfiram, yet another given a high-fat diet alongside a high dose of disulfiram, and a group given their standard diet. The mice were fed these arrangements for an additional 12 weeks.

The results were described as dramatic. The study found that overweight mice in the disulfiram groups experienced drops in both metabolic damage and body weight despite eating a high-fat diet. The mice given a higher dose of disulfiram lost up to 40-percent of their body weight in four weeks; mice in both of the drug groups also had improvements in blood glucose akin to the mice that were put on a standard diet without the medication.

NIA scientist Michel Bernier, Ph.D., explained:

When we first went down this path, we did not know what to expect, but once we started to see data showing dramatic weight loss and leaner body mass in the mice, we turned to each other and couldnt quite believe our eyes.

The study found that the benefits likely resulted from the drug directly, not from behavioral changes caused by the medication. The mice were not made to exercise as part of the study nor did the scientist notice any increased activity from them during the study. As well, the study didnt note any harm caused by the disulfiram use. Its important to remember, of course, that these results pertain to mice, not necessarily to humans.

Finding time to commit to exercise can be a real challenge, forcing many of us to squeeze in a quick run or gym session whenever there's a spare moment. But researchsuggests if you really want to make the most of your exercise, you should do it at the same time each day. It's a schedule your body will thank you for.

A 2019 study lead by researchers from Brown Alpert Medical School in the US showed it really doesn't matter if you're a dawn jogger or a twilight cyclist; it's the consistency that's key if it's weight-loss you're after.

The US Department of Health and Human Services suggests two and a half hours of moderate physical activity each week is the least we should be doing to keep healthy.

And not just a minute here or there, but at least 10 minutes of heart-pounding exercise in each session.

Needless to say, if you're fairly fit and in a good state of health, you're probably meeting this requirement. But many of those who have problems keeping their weight down often struggle to get the exercise they need.

Using survey results on the physical activity of 375 individuals exercising for weight loss, the researchers of the 2019 study identified a strong relationship between a moderate-to-vigorous level of exercise at the same time of day and the amount of time spent exercising.

Roughly half of the volunteers were morning people, which, when taken in context with a previous study by some of the same scientists, could indicate physical activity before you start your day is the way to go.

This preference for regularity might all come down to the way we think about our diary.

Activities we expect to do at set times such as picking up the kids, going to work, or attending social meetings aren't really things we give a great deal of conscious thought to.

This mindless repetition is referred to as automaticity in psychology circles, and has already been shown to be important when it comes to sticking to an exercise regime.

By actively considering how we could slot in quick walk or a treadmill session, we're more likely to cut back minutes of pulse-raising activity rather than commit to the exercise.

The secret is to therefore associate exercise with some pre-existing mental 'cue' for an appointment that you won't avoid, reducing the effort required if you had to plan an activity and then motivate yourself to see it through.

You might catch the 7:30 train, manage a short cardio workout at the gym near the office, and then be at your desk for that daily 9 am meeting. Or, if you're a night owl, going for a late run the moment you get home.

That walk to the train station might be a habit, but it doesn't count. Incidental exercise can be worked into a routine, but only if it's of a kind that makes your heart noticeably pump harder and demands effort.

"Repeatedly exercising in the presence of consistent cues, such as at the same time of day or in the same location, may help to establish cue exercise relationships," write the researchers.

On its own, a survey such as this can only go so far in demonstrating what causes something as complicated as an exercise habit. Individual motivators can't be dismissed, and more research is needed before any definitive claims can be made.

"It will also be important to determine whether there is a specific time of day that is more advantageous for individuals who have initial low physical activity levels to develop a physical activity habit," the study's first author, Leah Schumacher, said in 2019.

Around the globe, just under a third of women and nearly a quarter of all men aren't engaging in a level of physical activity that will keep them on the right side of healthy.

The reasons are no doubt complex and varied, and also seem to be linked with how much leisure time we actually have at out disposal.

It's hard, especially for busy folks, but getting your 150 minutes a week is important. Pick a time and stick to it.

Treatment with disulfiram, normally prescribed to treat alcohol use disorder, shows health benefits in animal study.

An off-label experiment in mice using disulfiram, which has been used to treat alcohol use disorder for more than 50 years, consistently normalized body weight and reversed metabolic damage in obese middle-aged mice of both sexes. The international study was led by researchers at the National Institute on Aging (NIA), part of the National Institutes of Health . The results were published online in the journal Cell Metabolism on May 14.

The scientific team studied groups of 9-month-old lab mice who had been fed a high-fat diet for 12 weeks. As expected, this diet made the mice overweight and they started to show signs of pre-diabetes-like metabolic problems, such as insulin resistance and elevated fasting blood sugar levels. Next, the scientists divided these mice into four groups to be fed four different diets for an additional 12 weeks: a standard diet alone, a high-fat diet alone, a high-fat diet with a low amount of disulfiram, or a high-fat diet with a higher amount of disulfiram. As expected, the mice who stayed on the high-fat diet alone continued to gain weight and show metabolic problems. Mice who switched to standard diet alone gradually saw their body weight, fat composition and blood sugar levels return to normal.

The mice in the remaining two groups, with either a low or high dose of disulfiram added to their still-fatty food, showed a dramatic decrease in their weight and related metabolic damage. Mice on the high disulfiram dose lost as much as 40% of their body weight in just four weeks, effectively normalizing their weight to that of obese mice who were switched back to standard diet. Mice in either disulfiram dose diet group became leaner and showed significant improvement in blood glucose levels on par with the mice who were returned to standard diet. Disulfiram treatment, which has few harmful side effects in humans, also appeared to protect the pancreas and liver from damage caused by pre-diabetic type metabolic changes and fat build up usually caused by eating a high-fat diet.

The NIA scientists, Michel Bernier, Ph.D., and Rafael de Cabo, Ph.D., collaborate frequently with researchers at NIH and beyond on studies into how changes in dietary patterns like intermittent fasting could lead to cognitive and physical health benefits. They first became interested in disulfiram after reading about the benefits this class of drug has shown in treating type 2 diabetes in rats, coupled with the growing interest in repurposing drugs that may also improve healthy aging.

When we first went down this path, we did not know what to expect, but once we started to see data showing dramatic weight loss and leaner body mass in the mice, we turned to each other and couldnt quite believe our eyes, Bernier said.

According to studys research team, the key to the positive results seem to stem from disulfirams anti-inflammatory properties, which helped the mice avoid imbalances in fasting glucose and protected them from the damage of fatty diet and weight gain while improving metabolic efficiency. Both groups of obese mice (control and disulfiram) were not subjected to any form of exercise, nor did they demonstrate noticeable spontaneous behavioral changes. Based on the evidence they observed, the researchers believe the beneficial results of disulfiram stem solely from the drug. They did not observe any negative side effects from disulfiram in the mice.

The research team stresses that these results are based on animal studies, and they cannot be extrapolated to any potential benefits for human at this point. It is recommended that disulfiram not be used off-label for weight management outside of the context of clinical trials. Still, given the findings, they are planning future steps for studying disulfirams potential, including a controlled clinical study to test if it could help individuals with morbid obesity lose weight, as well as deeper investigation into the drugs molecular mechanisms and potential for combining with other therapeutic interventions.
